Caracterización molecular de aislados de Staphylococcus aureus resistente a meticilina (SARM) utilizando mecA-PCR y PCR-RFLP

Se estudiaron 88 aislados clínicos de Staphylococcus aureus resistente a meticilina (SARM) recuperados por la red de vigilancia bacteriológica del Instituto Conmemorativo Gorgas de Estudios de la Salud del Laboratorio Central de Referencia en Salud Pública (ICGES-LCRSP) obtenidos en la República de Panamá con el objetivo de caracterizar molecularmente los aislados que circulan en nuestro país. Se amplificó el gen mecA y se demostró su presencia en el 100% de los aislados. La caracterización de los aislados se realizó mediante RFLP-PCR de los genes coa, spa, HVR y digestión con HaeII. Los aislados se agruparon en 5 categorías con base en los genes para los cuales se obtuvo amplificación. La categoría 1 con amplificación en los tres genes (24 aislamientos) generó 19 patrones de bandas diferentes. La categoría 2 con amplificación en los genes (coa y spa) agrupó 25 aislamientos con 17 patrones diferentes. La categoría 3 con amplificación en (coa y HVR) agrupó 10 aislamientos con 6 patrones. La categoría 4 con amplificación solo para coa con 11 aislamientos y 7 patrones. La categoría 5, sólo un aislamiento con amplificación para HVR generó solo un tipo de patrón. Los patrones de banda obtenidos son reproducibles y comparables con estudios previos. Nuestros resultados demuestran que RFLP-PCR es una metodología de escrutinio útil, fácil, rápida y discriminante para realizar vigilancia epidemiológica de infecciones causadas por el SARM.
